Several possibilities are available to sequence specific targets in a complex pool of DNA. Targeting specific sequences using amplicons is one option. Amplicons can be in the size range of direct sequencing or in the case of Long Range PCR, can be used as input material to construct small insert libraries.

As an alternative, Oxford Nanopore allows also Long Range -PCR amplicons to be sequenced directly. Oxford Nanopore has typically a higher sequencing error rate than Illumina sequencing.

Capturing with synthetic probes of the target is another, often used option. Capturing the complete coding part of a genome or exome capturing is a frequently used application.
